Netrin proteins are a family of laminin-related secreted proteins that provide guidance signals for axonal growth and cell migration during development. Netrin-1 is largely expressed in the developing nervous system and in mesodermal tissues. During embryonic development, netrin-1 diffuses through the neuronal epithelium, where it forms a chemoattractant gradient that directs axonal migration to the ventral midline of the spinal cord. Netrin-2 and the corresponding mouse homolog netrin-3 are expressed primarily in the lower two-thirds of the spinal cord, and, like netrin-1, they can either attract or repel commissural axons at a distance. High levels of netrin-4 mRNA have been detected in many cells and tissues, including cerebral cortex, hippocampus, amygdaloid nuclei and Purkinje cells. Netrin-4 is important in neural, kidney and vascular development. Netrin G1 and netrin G2 are both expressed in the brain but G1 is most predominantly expressed in the thalamus and G2 is most predominantly expressed in the cortex. These two proteins differ from classical netrins by their failure to bind netrin receptors, the presence of a glycosyl phosphatidylinositol membrane anchor and the generation of multiple isoforms.
