CD34 Antibody (43A1) is a mouse monoclonal IgG3 antibody that detects CD34 in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM). CD34 is a heavily glycosylated transmembrane glycoprotein that plays a crucial role in the regulation of hematopoietic stem cell adhesion and migration, making CD34 essential for the maintenance of stem cell niches and the proper functioning of the immune system. CD34 is predominantly expressed on the surface of lymphohematopoietic stem and progenitor cells, as well as small-vessel endothelial cells, embryonic fibroblasts, and certain cells in both fetal and adult nervous tissue. Notably, CD34 expression is highest in the most primitive stem cells, gradually diminishing as these cells differentiate into lineage-committed progenitors. The intracellular domain of CD34 contains consensus sites for phosphorylation by activated protein kinase C (PKC), along with serine, threonine, and tyrosine phosphorylation sites, which are critical for mediating signaling pathways that influence cell behavior. The presence of CD34 on capillary endothelial cells and bone marrow stromal cells further underscores CD34′s importance in vascular biology and tissue homeostasis. Anti-CD34 antibody (43A1) is an invaluable tool for researchers investigating stem cell biology, vascular development, and the mechanisms underlying hematopoiesis.
