Bcl-2 Antibody (C-2) is a mouse monoclonal IgG1 kappa light chain antibody that detects Bcl-2 protein of mouse, rat, and human origin through western blotting, immunoprecipitation, immunofluorescence, immunohistochemistry, and flow cytometry. Bcl-2 monoclonal antibody (C-2) is offered in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase, phycoerythrin, fluorescein isothiocyanate, and multiple Alexa Fluor® conjugates. Bcl-2 protein plays a crucial role as a key regulator of apoptosis, which is vital for normal development, maintaining tissue homeostasis, and providing defense against pathogens. Immunostaining of human tissues reveals that Bcl-2 is primarily localized in the cytoplasm and associated with the membrane, highlighting its function as an anti-apoptotic oncoprotein. Bcl-2 promotes cell survival by engaging in protein-protein interactions with other members of the Bcl-2 family, including the anti-apoptotic proteins Bcl-xL, Mcl-1, Bcl-w, and A1, as well as the pro-apoptotic proteins Bax, Bak, Bik, Bad, and Bid. Regulation of Bcl-2′s anti-apoptotic function can occur through mechanisms such as proteolytic processing and phosphorylation, which are essential for its activity. Additionally, Bcl-2 may inhibit the activation of the cytochrome c/Apaf-1 pathway by stabilizing the mitochondrial membrane, thereby preventing apoptosis. Mutations in the Bcl-2 gene have been implicated in various cancers, where normal apoptotic processes are disrupted due to the overexpression or deregulation of Bcl-2′s anti-apoptotic effects.
