Descripción
BCIP-NBT Solution is a 5-bromo, 4-chloro, 3-indolylphosphate (BCIP)/Nitro-Blue Tetrazolium (NBT) substrate, used for the localization of alkaline phosphatase (AP) labeled probes on western, northern, southern and dot blots. BCIP acts as the substrate for AP and reacts further after the dephosphorylation to give a dark-blue indigo-dye as an oxidation product; NBT serves herein as the oxidant and also gives a dark-blue dye, intensifying the color and making the detection more sensitive. Recommended for use with Crystal Mounting Medium (sc-24943).
Uso
• Product should be brought to room temperature (~25 °C) before use.
• Use enough substrate solution to completely cover the membrane surface.
• Upon incubation, a dark blue-purple reaction product will develop on membrane sites where alkaline phosphatase is present. Monitor reaction continuously and record results before background staining prevails.
• Stop the reaction by rinsing briefly with water.
• Dilution of the substrate is not recommended. To reduce the intensity of a reaction, it is recommended the antibodies or conjugates be diluted.
