MTA3 Antibody (428C2a) is a mouse monoclonal IgG1 kappa light chain antibody that detects MTA3 protein of human origin by western blotting (WB) and immunoprecipitation (IP). Anti-MTA3 antibody (428C2a) is a non-conjugated monoclonal isotype antibody that plays a crucial role in cancer biology. Metastasis-associated protein 3 (MTA3) is a vital subunit of the Mi-2/NuRD transcriptional corepressor complex, which is essential for regulating gene expression through the covalent modification of histone proteins. This regulation is particularly important in breast cancer, where MTA3 mediates the repression of the Snail transcription factor, helping maintain a differentiated epithelial status in cancer cells. MTA3 exists in two known isoforms: a short form that binds to estrogen receptors and sequesters them in the cytoplasm, enhancing non-genomic responses, and a long form that is localized in the nucleus. The differential localization and function of these isoforms underscore the complexity of MTA3′s role in cellular processes. MTA3 is widely expressed, with highest levels found in brain, adrenal glands, ovaries, and virgin mammary glands, and has been observed to be upregulated in tumors compared to adjacent normal tissues, highlighting potential as a biomarker for cancer progression.

I am using MTA3 (428C2a): sc-81325 antibody for immunoprecipitation, how much antibody should I use?

Thank you for your question. We recommend using 1–10 µl (i.e., 0.2–2 µg) of antibody with 1 ml of cell lysate. However, optimal antibody concentration should be determined by titration.
