SEE ALSO...

p53 Antibody (BP 53.12) is an IgG2a kappa light chain mouse monoclonal antibody, raised against the N-terminus of p53 of human origin, is an important tool used to detect and study the p53 protein within cells and tissues. This p53 mouse antibody (BP 53.12) specifically binds to p53, allowing researchers to examine its expression levels and localization through techniques such as ELISA, immunohistochemistry, western blotting, and immunofluorescence microscopy in human samples. Notably, it enables detection of both wild type and mutant p53 under denaturing and non-denaturing conditions. By utilizing the anti-p53 antibody (BP 53.12), scientists can investigate alterations in p53 that are common in various cancers, many of which involve mutations or dysregulation of this protein. Encoded by the TP53 gene, p53 protein is a fundamental tumor suppressor in human cells, acting as a transcription factor, p53 regulates genes that control vital cellular processes such as DNA repair, cell cycle arrest, programmed cell death, and senescence. When cells experience stress or DNA damage, p53 becomes activated and can pause the cell cycle at checkpoints like G1/S or G2/M, providing an opportunity for DNA repair mechanisms to fix the damage. If the damage is irreparable, p53 can initiate apoptosis to prevent the propagation of cells with genetic abnormalities, thereby inhibiting tumor development. Studying p53 with the help of p53 monoclonal antibody (BP 53.12) enhances our understanding of tumor suppression mechanisms and aids in the development of potential therapeutic strategies targeting the p53 pathway.

I am using p53 (BP 53.12): sc-81168 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Asked by: Randy McDonald
We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
