HCAM Antibody (DF1485) is a mouse monoclonal IgG1 kappa light chain antibody that detects HCAM protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and flow cytometry (FCM). Anti-HCAM antibody (DF1485) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. HCAM protein, also known as CD44, plays a critical role in cell adhesion and migration, functioning as a receptor for hyaluronic acid and interacting with ligands such as osteopontin. This interaction is vital for processes such as hematopoiesis, lymphocyte activation, and tumor metastasis, highlighting HCAM′s importance in both normal physiological functions and pathological conditions. HCAM is a 742 amino acid single-pass type I membrane protein that undergoes alternative splicing, resulting in multiple isoforms, including CD44R, CDw44, CD44S, CD44H, and CD44E. CD44H isoform is expressed at elevated levels in cancer tissues, suggesting significant involvement in tumor progression and making HCAM a potential target for therapeutic interventions. HCAM′s ability to mediate cell-cell and cell-matrix interactions underscores an essential role in embryogenesis and development, further emphasizing HCAM (DF1485) antibody′s relevance in research and clinical applications.

I would like more information on the antibody clone: which portions of the protein does it recognize? I am particularly interested in whether the antibody activates the receptor or not.

Thank you for your question. Anti-HCAM Antibody (DF1485): sc-7297 was raised against purified CD44 antigen (PGp-1) from lymphocyte membrane. No epitope mapping has been performed as of yet, so I am afraid we do not have any information on the exact epitope it recognises.

Is this antibody detecting all the Cd44 variants?

Thank you for your question. This anitbody was raised against the full length of the protein, we do not characterize the exact binding site of the antibody, and therefore we do not know if the antibody will detect all variants of CD44.

Which extraction procedure and inhibitors are recommended? I can't obtain the positive band of 90 kD in hole lysate and membrane fraction of CCRF-CEM cells using HCAM (sc7297) and m-IgGk BP-HRP (sc-516102) secondary AB.
