XPA Antibody (12F5) is a mouse monoclonal IgG2a kappa light chain antibody that detects XPA protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). XPA (12F5) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. XPA protein plays a critical role in the nucleotide excision repair (NER) pathway, which is essential for repairing DNA damage caused by ultraviolet (UV) light. This function is particularly important as deficiencies in NER can lead to xeroderma pigmentosum, a genetic disorder that significantly increases the risk of skin cancer due to the accumulation of unrepaired DNA lesions. XPA coordinates the assembly of the preincision complex necessary for recognizing and processing damaged DNA, and exhibits a unique ability to bind to both single-stranded and double-stranded DNA. XPA′s structure allows formation of a homodimer in the absence of DNA, enhancing binding efficiency when DNA is present. This dimerization is crucial for the effective repair of bulky DNA adducts, as XPA′s activity is regulated in a concentration-dependent manner, ensuring cells can respond appropriately to varying levels of DNA damage. Anti-XPA antibody (12F5) serves as a valuable tool for studying the mechanisms of DNA repair and the implications of its dysfunction in human health.
