SPOP Antibody (B-8) is a mouse monoclonal IgG2b kappa light chain antibody that detects SPOP protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-SPOP antibody (B-8) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. SPOP protein, also known as speckle-type POZ protein, TEF2, HIB homolog 1, or Roadkill homolog 1, plays a crucial role in cellular processes by mediating ubiquitinylation and protein degradation through interaction with the CUL-3 ubiquitin ligase complex. Structurally, SPOP contains an N-terminal MATH (Meprin and TRAF Homology) domain and a C-terminal BTB/POZ domain, which are essential for function; the MATH domain is responsible for substrate recognition, while the BTB/POZ domain facilitates interaction with CUL-3. This dual-domain architecture allows SPOP to effectively recruit various substrates, including PDX-1, Bmi-1, MacroH2A, PIPK II β, and Daxx, for ubiquitin-mediated degradation, thereby regulating critical cellular pathways. SPOP undergoes ubiquitination and subsequent proteasomal degradation, highlighting a dynamic role in maintaining protein homeostasis within the cell. Nuclear expression of SPOP across various tissues underscores importance in cellular regulation and potential implications in disease processes, making SPOP monoclonal antibody (B-8) a valuable tool for researchers studying these mechanisms.
