Insulin B Antibody (C-12) is a mouse monoclonal IgG1 kappa light chain antibody designed to target an epitope spanning amino acids 33-69 near the N-terminus of the human Insulin B chain. Insulin B monoclonal antibody (C-12) exhibits broad species reactivity, effectively recognizing insulin from mouse, rat, and human sources, as well as additional species including equine, canine, bovine, and porcine, making it versatile for various experimental applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-Insulin B antibody (C-12) is available in both non-conjugated form and multiple conjugated forms including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® dyes, providing flexibility for diverse research needs. Insulin itself is a critical secreted peptide hormone that regulates metabolic processes by increasing glucose uptake and glycogen synthesis, thereby reducing blood glucose levels. Insulin is synthesized initially as preproinsulin, which is subsequently processed to proinsulin and finally to the mature insulin hormone consisting of 51 amino acids divided into an A chain and a B chain connected by two disulfide bridges. Insulin enhances cellular permeability to monosaccharides, amino acids, and fatty acids, playing a vital role in maintaining energy homeostasis. Insulin is secreted by the pancreatic beta cells at basal levels and secretion significantly increases in response to elevated blood glucose levels. Insulin hormone exerts effects by binding to specific insulin receptors on target cell membranes, triggering signaling pathways that facilitate glucose uptake and utilization. Proper insulin function is essential for preventing metabolic disorders, and defects in insulin production or signaling are major causes of hyperproinsulinemia and type-II diabetes mellitus. By providing precise detection of the Insulin B protein, Insulin B (C-12) antibody is an indispensable tool for researchers studying insulin′s role in metabolism, regulatory mechanisms, and involvement in various pathological conditions.

Can I use SC-377071 for sandwich ELISA?

Thank you for your question. This antibody, sc-377071 Insulin B (C-12), has not been tested for sandwich ELISA, but it is recommended and warrantied for direct or indirect ELISA.

The expected molecular weight of Insulin is 6-7 kDa. Why 12 kDa? It´s not clear. Thank you.

Thank you for your question. The predicted molecular weight of 12 kDa is for the precursor of Insulin B which is 110 amino acids in length.

How can Insulin B Antibody (C-12): sc-377071 be used for double or triple staining, if the other primary antibodies are also raised in mouse?

In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This mouse monoclonal antibody is currently available conjugated to phycoerythrin (sc-377071 PE), fluorescein (sc-377071 FITC), Alexa Fluor® 488 (sc-377071 AF488) or Alexa Fluor® 647 (sc-377071 AF647).

Wide-field diffuse amacrine cells in the monkey retina contain immunoreactive Cocaine- and Amphetamine-Regulated Transcript (CART). Peptides. 2016 Oct;84:22-35). The isolated retina of a baboon was fixed for 60 minutes in 4% paraformaldehyde. Tissue was incubated for 9 days in a 1:1,000 solution of primary antibody with detergent and a blocking agent at 4 °C, washed, then incubated in an affinity-purified secondary antibody for 4 days at 4 °C, washed, and mounted in medium containing DAPI. No peptide blocking control was performed. Results: Very light punctate labeling and low background throughout the optic fiber layer, ganglion cell layer, inner plexiform layer, and inner nuclear layer (less).
