Cathepsin D, also known as CTSD, is a lysosomal aspartyl protease involved in protein degradation and the processing of protein precursors into bioactive peptides. This protein is synthesized as an inactive precursor that undergoes proteolytic cleavage to generate an active enzyme, a post-translational modification essential for activity within the acidic lysosomal environment. Glycosylation further stabilizes cathepsin D and facilitates proper trafficking to lysosomes. Cathepsin D plays a crucial role in cellular processes such as autophagy, apoptosis, and immune regulation. Dysregulation of cathepsin D activity or post-translational modifications has been associated with diseases including cancer, where cathepsin D contributes to tumor invasion and metastasis, and neurodegenerative disorders. The gene encoding cathepsin D is tightly regulated by transcription factors such as Sp1 and AP-2, reflecting its importance in maintaining cellular homeostasis.
