Description
- A protein kinase C (PKC) inhibitor which also serves as an intracellular Ca²⁺ antagonist. TMB-8 possesses both Ca²⁺ channel- and Na₊ channel-blocking properties and reduces the membrane K⁺ conductance. Shown to nonselectively inhibit membrane currents in single isolated guinea pig ventricular cardiomyocytes and to inhibit 5-hydroxytryptamine type 3A (5-HT3A, SR-3A) receptor-mediated currents. TMB-8 inhibits platelet-activating factors (PAF) and PMA-stimulated prostaglandin production in Kupffer cells. Research shows that TMB-8 accumulates at the lipid-water interphase of biologic membranes and therefore interferes with the normal function of many membrane proteins. A useful tool to distinguish dependence on intracellular versus extracellular Ca²⁺.
