E-cadherin Antibody (67A4) is a mouse monoclonal IgG1 kappa light chain antibody that detects human E-cadherin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and flow cytometry (FCM). Anti-E-cadherin antibody (67A4) is available in non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. E-cadherin, a member of the cadherin family, plays a crucial role in cell-cell adhesion, essential for maintaining tissue integrity and facilitating morphogenetic processes during development. E-cadherin primarily localizes at the cell membrane, forming adherens junctions that connect adjacent cells and contribute to tissue structural stability. This membrane localization enables E-cadherin to mediate strong intercellular adhesion, necessary for epithelial layer formation and prevention of invasive behaviors in cancer cells. E-cadherin interaction with cytoplasmic proteins, such as β-catenin, is critical for signaling pathways that regulate cell proliferation and differentiation, highlighting its importance in both normal physiology and disease states.

Rated 1 out of 5 by from Anticorpo non funzionantePrimary antibodies E-Cadherin (67A4) sc-21791 was used diluted 1:500 according to the manufacturer’s recommendations (for Western Blotting dilution range 1:100-1:1000), in Milk 5%. Membrane of PVDF has been incubated overnight at 4°C and then it has been probed for 1h at RT with Santa Cruz secondary antibodies diluted 1:5000 in Milk 1%. We have tested this antibody on two different sets of samples: in six cancer cell lines and in human activated PBMCs. Moreover, even if we tested the antibody at higher concentration, we only obtained a higher background but not an increase in the specific protein signal; therefore we show only the blots obtained after the incubation with antibody prepared as described above. In conclusion: • E-Cadherin did not work at all, neither after a long exposure. We do not have tools to verify the specificity for this antibody too. All the membranes have been probed with a normalizer and each gel has been stained with Coomassie Brillant Blue, shown on the right side of each panel, in order to demonstrate that the samples ran well and that the proteins are present on the SDS-PAGE.
