T-bet Antibody (4B10) is a mouse monoclonal IgG1 kappa light chain antibody that detects T-bet protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), flow cytometry (FCM), and enzyme-linked immunosorbent assay (ELISA). T-bet (4B10) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. T-bet protein, also known as TBX21, plays a crucial role in the differentiation of T helper lymphocytes into Th1 cells, which are essential for immune response against intracellular pathogens. T-bet is a T box transcription factor that promotes expression of Th1 cytokine interferon-gamma (IFN-γ) while inhibiting development of Th2 cells, maintaining balance in immune response. This regulatory function helps prevent excessive Th2 responses that can lead to allergic reactions and asthma. T-bet is selectively expressed in Th1 cells, with expression upregulated by signals from T cell receptor and interleukin-12, highlighting importance in T cell activation and differentiation. Originally isolated from nuclear extracts of resting and PMA/ionomycin-activated AE7 cells, T-bet is found at low levels in resting cells but increases significantly upon stimulation, underscoring role in adaptive immune response.
