Meclofenamate sodium is a dual inhibitor of the 5-lipoxygenase (5-LO) and cyclooxygenase (COX) pathways. Meclofenamate sodium is demonstrated to inhibit both COX-1 and COX-2, showing micromolar inhibition of human recombinant hCOX-1 (IC50 = 1.5 μM) and hCOX-2 (9.7 μM). An irreversible time-dependent inhibition of hCOX-2 is described, involving a rapid equilibrium binding of enzyme and inhibitor followed by formation of a tightly bound inhibited complex.
