p-MYL9 Background Information ROCK (Rho-kinase), an effector molecule of RhoA, phosphorylates the myosin binding subunit (MBS) of myosin phosphatase to inhibit the intrinsic phosphatase activity (1–3). This inhibition increases phosphorylation of myosin light chain (MLC) of myosin II, which is suggested to induce RhoA-mediated assembly of stress fibers and focal adhesions (1). Increases in cytosolic Ca2+ concentrations and phosphorylation of MLC are key steps of normal and abnormal contractility in smooth muscle (4). In a pathological condition like vasospasm, the coronary artery exhibits a hyperreactive contraction associated with an enhanced and sustained MLC phosphorylation, particularly diphosphorylation (Threonine 18/Serine 19) (2,4–7). The phosphorylation level is regulated by a balance between phosphorylation and dephosphorylation (4). Myosin phosphatase activity is regulated through the phosphorylation of MBS130K of the myosin phosphatase in smooth muscle (5).
