P-Selectin Antibody (1 E 3) is a mouse monoclonal IgG2a kappa light chain antibody that detects P-Selectin of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM). P-Selectin (1 E 3) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. P-Selectin, also known as GMP-140 or CD62P, plays a crucial role in mediating the adhesion of platelets and leukocytes to the endothelium during inflammatory responses, which is vital for the recruitment of immune cells to sites of injury or infection. P-Selectin is primarily located on the surface of activated platelets and endothelial cells, where P-Selectin facilitates the initial stages of cell adhesion and migration. Proper functioning of P-Selectin is essential for maintaining vascular integrity and regulating immune responses, making P-Selectin a significant target for research in cardiovascular diseases and inflammatory conditions. P-Selectin interacts with specific carbohydrates on neutrophils and monocytes, and P-Selectin expression is upregulated in response to inflammatory stimuli, highlighting P-Selectin′s importance in the pathophysiology of various diseases.

Regarding my question about another antibody, I'm not looking for a secondary antibody, but for another P-selectin antibody that could be used as the detector in ELISA that would bind at a spatially distinct epitope. Can you recommend one? Thanks

Thank you for your question. We do not characterize pairs of monoclonal antibodies for use in sandwich ELISA. Many of our antibodies are recommended as detection antibodies in direct or indirect ELISA, however we cannot recommend a matching capture antibody. Do you have any data on performance in ELISA? If the antibody is known or predicted to work in ELISA, can you recommend a second antibody to use in the ELISA pair? Thank you

Thank you for your question. We have not yet tested this specific mouse monoclonal, P-Selectin Antibody (1 E 3): sc-19672, for ELISA so we do not have any specific recommendations we can offer at this time for this research application. However, the antibody has been tested in Western blot, immunohistochemistry and flow cytometry so we believe it should also work for ELISA. P-Selectin Antibody (1 E 3) is a mouse monoclonal IgG2a, so for a secondary antibody you would want to use an anti-mouse IgG secondary antibody conjugated to either alkaline phosphatase (AP) or horseradish peroxidase (HRP), depending on what detection method being used. We would recommend our mouse IgG binding protein conjugated to HRP, m-IgG kappa BP-HRP: sc-516102. Can P-Selectin (1 E 3): sc-19672 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, P-Selectin (1 E 3): sc-19672 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Thank you for your question. We do not know whether or not this antibody will detect canine. P-Selectin Antibody (1 E 3) is a mouse monoclonal raised against fixed thrombin-activated human platelets. The exact epitope recognized by this antibody has not been determined so we are unable to try to predict cross-reactivity based on sequence analysis. This would have to be determined experimentally.
