JAM-A Background Information Junctional adhesion molecule (JAM) is a member of the immunoglobulin superfamily expressed in tight junctions of epithelial cells and endothelial cells, and it is implicated in transendothelial migration of leukocytes. JAM is constitutively expressed on circulating monocytes, neutrophils, lymphocytes subsets, and platelets. JAM changes its distribution in response to proinflammatory cytokines. The JAM family consists of JAM-1 and JAM-2. JAM-1 localizes with F-actin at the cell-cell contacts and at the membrane ruffles, but not at the stress fibers, and is involved in cell to cell, adhesion through homophilic interactions. The PDZ domain protein AF-6 is an intracellular binding partner of JAM-1, which is recruited to intercellular junctions by this interaction with AF-6 and possibly with ZO-1. JAM-1 also binds directly to s1 and permits reovirus-induced activation of NF-kB and apoptosis. JAM-1 plays a role in the organization of tight junctions and modulates leukocyte extravasation through endothelial intercellular junctions in vitro and in vivo. JAM-1 is able to homodimerize and form clusters at several sites of cell-cell contact in aggregated platelets. Upon platelet activation, JAM-1 is phosphorylated by protein kinase C at serine 284. |
