ESET Antibody (A-1) is a mouse monoclonal IgG2b kappa light chain antibody that detects ESET protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-ESET antibody (A-1) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The ERG-associated protein with SET domain (ESET), also known as Histone H3-K9 methyltransferase 4 or SET domain bifurcated 1, is a crucial nuclear protein that belongs to the histone-lysine methyltransferase family and the Suvar3-9 subfamily. This highly conserved protein, consisting of 150 amino acids, plays a significant role in chromatin structure modulation, which is essential for regulating gene expression and maintaining cellular identity. ESET is specifically localized to nonpericentromeric regions of chromatin and is excluded from cell nucleoli and areas of condensed chromatin, highlighting its importance in epigenetic regulation. The gene encoding ESET, SETDB1, is located on chromosome 1q21, and mutations in this gene can disrupt methyltransferase activity, leading to altered gene expression patterns. ESET functions by methylating Lys-9 of histone H3, a modification that serves as a signal for epigenetic transcriptional repression by recruiting heterochromatin protein 1 (HP1) to methylated histones. ESET is widely expressed, with highest levels found in testis, underscoring its potential role in reproductive biology and development.

What kind of blocking solution do you recommend and which dilution? Antibody is still not working on nuclear extracts.

Thank you for your question. sc-166621 is recommended to be used in a dilution range of 1:100-1:1000. We recommend using TBS Blotto A: sc-2333 or TBS Blotto B: sc-2335 for blocking your western blot membranes.
