HFE plays a crucial role in iron homeostasis by regulating iron absorption in the intestine, which is vital for preventing iron overload conditions such as hereditary hemochromatosis. HFE is primarily located in the crypt enterocytes of the duodenum, where HFE exhibits a unique intracellular localization essential for function. In the placenta, HFE co-localizes with the transferrin receptor (TfR), forming a stable association that facilitates iron transport.
