HFE Antibody (44-H) is a mouse monoclonal IgG1 antibody that detects HFE in human samples through applications such as western blotting (WB), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). HFE plays a crucial role in iron homeostasis by regulating iron absorption in the intestine, which is vital for preventing iron overload conditions such as hereditary hemochromatosis. HFE is primarily located in the crypt enterocytes of the duodenum, where HFE exhibits a unique intracellular localization essential for function. In the placenta, HFE co-localizes with the transferrin receptor (TfR), forming a stable association that facilitates iron transport. The interaction between HFE and TfR helps modulate iron uptake and release, maintaining iron balance in the body. Immunocytochemistry has demonstrated that both HFE and TfR are expressed in crypt enterocytes, while western blotting (WB) has confirmed their physical association with β2-microglobulin. HFE has two mutually exclusive activities: inhibiting iron uptake or inhibiting iron release, with the balance between serum transferrin saturation and serum transferrin receptor concentrations determining which function predominates. The gene encoding HFE is located on human chromosome 6p21.3, highlighting genetic significance in iron metabolism disorders.
