p-eIF2α Background Information Phosphorylation of the a subunit of eukaryotic initiation factor 2 (eIF2å) is known to be an important translational control mechanism (1). Regulation of mammalian eIF2å activity is directly governed by specific phosphorylation on Serine 52 (1). In mammalian cells, eIF2å is phosphorylated at Serine 52 by at least two kinases, the heme-controlled repressor and the interferon-inducible double stranded RNA-dependent protein kinase (2,3). This phosphorylation event results in an inhibition in the exchange of bound nucleotides and the recycling of eIF2å (4–6). In addition, phosphorylation of eIF-2å blocks the GDP-GTP exchange activity of eIF2∫, resulting in the suppression of protein synthesis (2,3).
